    Abstractions and Algorithms for Control of Extensible and Heterogeneous Virtualized Network Infrastructures

    Virtualized network infrastructures are currently deployed in both research and commercial contexts. The complexity of the virtualization layer varies greatly in different deployments, ranging from cloud computing environments, to carrier Ethernet applications using stacked VLANs, to networking testbeds. In all of these cases, many users are sharing the resources of one provider and each user expects their resources to be isolated from all other users. There are many challenges associated with the control and management of these systems, including resource allocation and sharing, resource isolation, system security, and usability. Among the different types of virtualized infrastructures, network testbeds are of particular interest due to their widespread use in education and in the networking research community. Networking researchers rely extensively on testbeds when evaluating new protocols and ideas. Indeed, a substantial percentage of top research papers include results gathered from testbeds. Network emulation testbeds in particular are often used to conduct innovative research because they allow users to emulate diverse network topologies in a controlled environment. That is, researchers run experiments with a collection of resources that can be reconfigured to represent many different network scenarios. The user typically has control over most of the resources in their experiment which results in a high level of reproducibility. As such, these types of testbeds provide an excellent bridge between simulation and deployment of new ideas. Unfortunately, most testbeds suffer from a general lack of resource extensibility and diversity. This dissertation extends the current state of the art by designing a new, more general testbed infrastructure that expands and enhances the capabilities of modern testbeds. This includes pertinent abstractions, software design, and related algorithms. The design has also been prototyped in the form of the Open Network Laboratory network testbed, which has been successfully used in educational and research pursuits. While the focus is on network testbeds, the results of this research will also be applicable to the broader class of virtualized system infrastructures

    Phorbol myristate acetate and Bryostatin 1 rescue IFN-gamma inducibility of MHC class II molecules in LS1034 colorectal carcinoma cell line

    BACKGROUND: The expression of major histocompatibility complex class II (MHCII) antigens in both mouse and human tumors is rare, and these antigens are not easily inducible by IFN-gamma (IFNg). Since MHCII may play an important role in the development of host antitumor immune response, we explored the possibility of restoring MHCII inducibility in several IFNg-resistant tumor cell lines using protein kinase C (PKC) agonists phorbol myristate acetate (PMA) or Bryostatin. RESULTS: Tumor cells were co-cultured with various concentrations of PMA and IFNg for 48 hr. The expression of MHCII antigens and receptors IFNgR1 and IFNgR2 was determined by flow cytometry. We showed that the presence of as little as 0.1 ng/ml of PMA in tissue culture restored the ability of weakly inducible LS1034 colon carcinoma cells to express MHCII in response to IFNg (100 – 10,000 IU/ml) in a dose-dependent manner. Likewise, Bryostatin 1, as low as 10 ng/ml produced a 5–6 fold upregulation of MHCII. The effect of PMA was not observed in two other poorly responding cell lines, MSTO-211H mesothelioma and HepG2 hepatocellular carcinoma, and was abrogated by relatively high concentrations of PKC inhibitors staurosporine (100 nM) and GF 109203X (1,000 nM). Both surface and intracellular staining of all cell lines with antibodies against IFNgR1 and IFNgR2 failed to detect any increase in IFNg receptor expression following incubation with PMA. CONCLUSION: In this study we showed that IFNg-inducibility of MHCII antigens in weakly inducible LS1034 colorectal carcinoma cell line can be rescued by concomitant incubation with PKC agonists. Bryostatin 1 may be considered for further investigation of IFNg-dependent MHCII induction in resistant tumors in vivo

    Weak measurement and rapid state reduction in bipartite quantum systems

    In this paper we consider feedback control algorithms for the rapid purification of a bipartite state consisting of two qubits, when the observer has access to only one of the qubits. We show 1) that the algorithm that maximizes the average purification rate is not the same as that that for a single qubit, and 2) that it is always possible to construct an algorithm that generates a deterministic rate of purification for {\em both} qubits. We also reveal a key difference between projective and continuous measurements with regard to state-purification.Comment: 4 pages, 3 figure

Dynamic feedback loops between self and other, cause and effect, represented and representamen may no longer seem surprising; but, in spite of our enhanced awareness, little deliberate attention is devoted to modeling or grounding such phenomena. Attending to both linguistic and extra-linguistic modalities of chiasmus (the X figure), a group of anthropologists has recently embraced this challenge. Applied to contemporary problems in linguistic anthropology, chiasmus functions to highlight and enhance relationships of interdependence or symbiosis between contraries, including anthropology’s four fields, the nature of human being and facets of being human

    Frequency Tracking and Parameter Estimation for Robust Quantum State-Estimation

    In this paper we consider the problem of tracking the state of a quantum system via a continuous measurement. If the system Hamiltonian is known precisely, this merely requires integrating the appropriate stochastic master equation. However, even a small error in the assumed Hamiltonian can render this approach useless. The natural answer to this problem is to include the parameters of the Hamiltonian as part of the estimation problem, and the full Bayesian solution to this task provides a state-estimate that is robust against uncertainties. However, this approach requires considerable computational overhead. Here we consider a single qubit in which the Hamiltonian contains a single unknown parameter. We show that classical frequency estimation techniques greatly reduce the computational overhead associated with Bayesian estimation and provide accurate estimates for the qubit frequencyComment: 6 figures, 13 page
